ST2 is a member of the Toll-like receptor/interleukin-1 (interleukin-1, IL-1) receptor superfamily. IL-33 is its specific functional ligand and is secreted by cardiomyocytes and fibroblasts. Two products of gene expression: transmembrane ST2 (ST2L) and sST2. ST2L contains three extracellular immunoglobulin domains, while sST2 lacks transmembrane and intracellular receptor domains. They bind to the common ligand IL-33 and play a biological role. ST2L and IL-33 signaling pathway has cardioprotective effects such as anti-cardiomyocyte hypertrophy, myocardial fibrosis and anti-atherosclerosis. When the heart load increases, sST2 secretion increases, and the increased sST2 prevents IL-33 from combining with ST2L, thereby resisting the cardioprotective effect of IL-33/ST2L signaling pathway. It is inferred that sST2 may be a pathogenic mediator of cardiomyocyte hypertrophy and myocardial fibrosis. Quantitative determination of sST2 levels can provide doctors with an accurate tool to assist in the assessment of heart failure.
